DUSP2 Deletion Inhibits Macrophage Migration by Inhibiting ERK Activation in Zebrafish

چکیده
Dual-specificity phosphatase 2 (DUSP2) regulates the activation of members mitogen-activated protein kinase (MAPK) family, which is involved in a variety cellular processes including cell proliferation, differentiation, apoptosis, and migration. DUSP2 also expression inflammatory mediators macrophages; however, it remains unknown whether participates macrophage Here, using tail fin injury model zebrafish larvae, we found that deletion inhibited pro-inflammatory cytokines chemokines. Moreover, live imaging results showed migration macrophages to site was after deletion. This inhibitory effect mediated through reduced extracellular regulated kinases (ERK) knockout zebrafish.

Matrix metalloproteinase-28 deletion exacerbates cardiac dysfunction and rupture after myocardial infarction in mice by inhibiting M2 macrophage activation.
RATIONALE Matrix metalloproteinase (MMP)-28 regulates the inflammatory and extracellular matrix responses in cardiac aging, but the roles of MMP-28 after myocardial infarction (MI) have not been explored. OBJECTIVE To determine the impact of MMP-28 deletion on post-MI remodeling of the left ventricle (LV). METHODS AND RESULTS Adult C57BL/6J wild-type (n=76) and MMP null (MMP-28((-/-)), n=86...
متن کاملInhibiting the HSP90 chaperone destabilizes macrophage migration inhibitory factor and thereby inhibits breast tumor progression
Intracellular macrophage migration inhibitory factor (MIF) often becomes stabilized in human cancer cells. MIF can promote tumor cell survival, and elevated MIF protein correlates with tumor aggressiveness and poor prognosis. However, the molecular mechanism facilitating MIF stabilization in tumors is not understood. We show that the tumor-activated HSP90 chaperone complex protects MIF from deg...
متن کاملNorepinephrine Inhibits Macrophage Migration by Decreasing CCR2 Expression
Increased incidences of infectious and septic complications during post-burn courses represent the main contributor to burn injury mortality. Sustained increases in catecholamine levels, especially norepinephrine (NE), contribute to immune disturbances in severely burned patients. The precise mechanisms underlying NE-mediated immunoregulation are not fully understood. Here we hypothesize that p...
متن کاملAtorvastatin inhibits insulin synthesis by inhibiting the Ras/Raf/ERK/CREB pathway in INS-1 cells
BACKROUND Type 2 diabetes has become a global epidemic disease. Atorvastatin has become a cornerstone in the prevention and treatment of atherosclerosis. However, increasing evidence showed that statins can dose-dependently increase the risk of diabetes mellitus. The mechanism is not clear. OBJECTIVE The Ras complex pathway (Ras/Raf/extracellular signal-regulated kinase [ERK]/cAMP response el...
متن کاملExtracellular pressure stimulates macrophage phagocytosis by inhibiting a pathway involving FAK and ERK.
We hypothesized that changes in extracellular pressure during inflammation or infection regulate macrophage phagocytosis through modulating the focal adhesion kinase (FAK)-ERK pathway. Undifferentiated (monocyte-like) or PMA-differentiated (macrophage-like) THP-1 cells were incubated at 37 degrees C with serum-opsonized latex beads under ambient or 20-mmHg increased pressure. Pressure did not a...
